The separation principle of silica gel chromatography is to separate substances according to their different adsorption forces on silica gel. Generally, substances with higher polarity are easily adsorbed by silica gel, and substances with weaker polarity are not easily adsorbed by silica gel. The whole chromatography process is the process of adsorption, desorption, re-adsorption, and re-desorption.


According to different filling matrix and sample distribution exchange principle, ion exchange chromatography, gel filtration chromatography and affinity chromatography are three classical chromatography techniques for separating mixtures.
